In vitro study | MCC950 inhibits classical and non-classical NLRP3 activation with IC50 of 7.5 and 8.1 nM in bone marrow-derived macrophages and human monocyte-derived macrophages, respectively. MCC950 specifically inhibits the activation of NLRP3 and does not inhibit AIM2, NLRC4 or NLRP1 These inflammasomes. |
In vivo study | MCC950 reduces interleukin-1β (IL-1β) production and reduces the severity of experimental autoimmune encephalomyelitis. In a mouse model of the autoinflammatory disease Cryopyrin-associated cyclic syndrome, MCC950 was able to rescue neonatal death. In indirect in vitro experiments, it was active in samples derived from patients with Veyer's syndrome. |
